Updated Functional Classification of -Lactamases

چکیده

Two classification schemes for -lactamases are currently in use. The molecular classification is based on the amino acid sequence and divides -lactamases into class A, C, and D enzymes which utilize serine for -lactam hydrolysis and class B metalloenzymes which require divalent zinc ions for substrate hydrolysis. The functional classification scheme updated herein is based on the 1995 proposal by Bush et al. (K. Bush, G. A. Jacoby, and A. A. Medeiros, Antimicrob. Agents Chemother. 39:1211–1233, 1995). It takes into account substrate and inhibitor profiles in an attempt to group the enzymes in ways that can be correlated with their phenotype in clinical isolates. Major groupings generally correlate with the more broadly based molecular classification. The updated system includes group 1 (class C) cephalosporinases; group 2 (classes A and D) broad-spectrum, inhibitor-resistant, and extended-spectrum -lactamases and serine carbapenemases; and group 3 metallo-lactamases. Several new subgroups of each of the major groups are described, based on specific attributes of individual enzymes. A list of attributes is also suggested for the description of a new -lactamase, including the requisite microbiological properties, substrate and inhibitor profiles, and molecular sequence data that provide an adequate characterization for a new -lactam-hydrolyzing enzyme.

Network Analysis of Sequence-Function Relationships and Exploration of Sequence Space of TEM β-Lactamases.

The Lactamase Engineering Database (www.LacED.uni-stuttgart.de) was developed to facilitate the classification and analysis of TEM β-lactamases. The current version contains 474 TEM variants. Two hundred fifty-nine variants form a large scale-free network of highly connected point mutants. The network was divided into three subnetworks which were enriched by single phenotypes: one network with ...

Characterization of β-Lactamases from Urinary Isolates of Escherichia coli in Tehran

Knowledge of antimicrobial resistance patterns in E. coli, the predominant pathogen associated with urinary tract infections (UTI) is important as a guide in selecting empirical antimicrobial therapy. Methods: To describe the antimicrobial susceptibility of E. coli associated with UTI in a major university hospital in Tehran (Iran), seventy-six clinical isolates of E. coli were studied for susc...

[The widening challenge of extended spectrum β-lactamases].

Since 1980, the prescription of new semi-synthetic molecules of third generation cephalosporins changed the course of modern medicine. However, the acquired resistance against these antibiotics was rapidly developed with the production of extended spectrum β-lactamases (ESBL) (TEM and SHV types) disseminated mainly by nosocomial Klebsiella pneumoniae clones. Since around 2000, we are facing a w...

Increasing the accuracy of the classification of diabetic patients in terms of functional limitation using linear and nonlinear combinations of biomarkers: Ramp AUC method

The Area under the ROC Curve (AUC) is a common index for evaluating the ability of the biomarkers for classification. In practice, a single biomarker has limited classification ability, so to improve the classification performance, we are interested in combining biomarkers linearly and nonlinearly. In this study, while introducing various types of loss functions, the Ramp AUC method and some of...
